PROTEIN ASSAY
TITLE : The comparison of protein concentration in fresh water between marine fishes by using Biuret and Lowry assays
OBJECTIVES :
To determine the concentration of protein between freshwater and marines fishes
To determine protein concentration using two types of protein assays which are Biuret and Lowry assays.
METHOD :
FINDINGS :
As the finding, the concentration of protein in marine water is higher than in freshwater. the protein reading in mackerel fish in marine water is highest which is 6.3694 mg/ml by using Biuret Method. Meanwhile,by using the Lowry Method, the reading was recorded by 6.41728 mg/ml. In the other hand for talapia which is freshwater which compared to marine water. The protein concentration was recorded as 0.8529 mg/ml for Biuret Method . Furthermore, by using Lowry Method was recorded as 1.3100 mg/mL .
WHAT I'VE LEARNT :
There are many things and techniques that I have learnt from this experiment. First of all, I learn how to measure the protein concentration of fish using two methods, which is Biuret and Lowry Method. Thi smethod is very useful for me to get an accurate level of protein concentration in each fish species. We are able to know which type of fish has the highest protein. Other that that, I also learn how to use new tool such as spectrophometer. The spectrophotometer is an instrument which measures the amount of light of a specificed wavelength which passes through a medium. According to Beer's law, the amount of light absorbed by a medium is proportional to the concentration of the absorbing material or solute present.

METHOD :
- The fish sample is weighted 20 grams and macerated into smaller size.
- We blended the fish in Phosphate Buffer Saline at 1:10 ratio.
- We incubated it for 24hours at 42 degree celcius in 100rpm shaker. the sample is centrifuged at 4 degree celcius, 1400 rpm for 15 minutes.
- The supernatant is collected and filtered again using Whitman filter.
- The protein content is measured using Lowry or Biuret method.
- We measured the absorbance at 750 nm and standard curve is plotted.
